Methanbildung ist die mikrobielle Bildung von Methan (CH4). Sie stellt die letzte Stufe des anaeroben Abbaus von biogenen Stoffen unter anoxischen Bedingungen dar. In dieser obligat anaerob ablaufenden Phase wird Essigsäure, die während der Acidogenese und Acetogenese gebildet wurde, durch acetoklastische (Essigsäure abbauende) Methanbildner in Methan umgewandelt. Kohlenstoffdioxid (CO2) und elementarer Wasserstoff (H2) werden durch hydrogenotrophe (sich von Wasserstoff ernährende) Methanbildner in Methan und Wasser umgewandelt.

Acetoklastische Methanbildung:

\mathrm{CH_3COOH \rightarrow CO_2 + CH_4 }

Methanbildung aus Kohlenstoffdioxid und elementarem Wasserstoff:

\mathrm{CO_2 + 4 \ H_2 \rightarrow CH_4 + 2 \ H_2O}

Bei dieser Reduktion von Carboxylgruppen (-COOH) zu Methan und Kohlenstoffdioxid und der Reduktion von Kohlenstoffdioxid zu Methan spielen Enzyme mit speziellen, charakteristischen Coenzymen eine Rolle, die nur bei Methanbildnern vorkommen. Insbesondere sind dies die Coenzyme Tetrahydromethanopterin, Coenzym M und ein Corrin-Enzym, sowie spezielle Elektronen- bzw. Wasserstoffüberträger.

Diese Umsetzungen sind exergon, das heißt, es wird dabei Energie frei. Sie dienen den Methanbildnern als Energiequelle. Die Änderung der Freien Energie (ΔG0') unter Standardbedingungen, jedoch bei pH = 7, beträgt bei der Methanbildung aus Essigsäure − 36 kJ mol−1 und bei der Methanbildung aus Kohlenstoffdioxid und Wasserstoff − 131 kJ mol−1 [1]. .
