Amylopektin ist der Hauptbestandteil (70–80 %) der natürlichen pflanzlichen Stärke, z. B. der Mais- oder der Kartoffelstärke. Den zweiten Hauptanteil bildet mit 20–30 % das Polysaccharid Amylose.

Das Polysaccharid Amylopektin hat eine Molare Masse von 200.000–1.000.000 g·mol-1,[1] entsprechend etwa 1.200–6.200 D-Glucose-Monomeren, die α-1,4-glykosidisch miteinander verbunden sind. Etwa alle 25 Monomere erfolgt eine α-1,6-glykosidische Verknüpfung, wodurch eine baumartige Verzweigung entsteht. Aufgrund dieses Unterschiedes in der Struktur bildet Amylopektin auf molekularer Ebene Knäuel-, bzw. Verzweigte Strukturen aus, die Verzweigungspunkte liegen hauptsächlich am C6 Atom. Amylose formt hingegen lineare Ketten.

Bei der Iodprobe mit Iod-Kaliumiodid-Lösung (Lugolsche Lösung) ergibt sich eine Rotfärbung.
