Chymosin ist eine Aspartat-Endopeptidase (Protease). Rinder-Chymosin findet sich als Labferment im vierten Magen (Labmagen) von Kälbern. Es wird von den Zellen der Magenschleimhaut in Form des Vorläuferproteins PreProChymosin produziert und als ProChymosin ins Lumen sezerniert. In der sauren Umgebung des Magens konvertiert das inaktive Zymogen ProChymosin zum aktiven Enzym Chymosin.

Struktur

Das bovine Protein Chymosin besteht aus 323 Aminosäuren, mit einer Molekülmasse von 35.653 Da.[1]

Es kommt in zwei Formen vor, die sich in ihrer Primärstruktur an Aminosäureposition 244 (Asparaginsäure oder Glycin) unterscheiden. Die A-Form hat eine leicht höhere spezifische Aktivität für κ-Casein, ist jedoch marginal weniger stabil als die B-Form.

Verwendung

Chymosin wird zur Spaltung von Milcheiweiß bei der Produktion von Käse eingesetzt, wobei es auch gentechnisch hergestellt wird.[2]
