John James Rickard Macleod (* 6. September 1876 in Cluny bei Dunkeld, Perthshire, Schottland; † 16. März 1935 in Aberdeen, Schottland) war ein schottisch-kanadischer Physiologe.

John Macleod studierte Medizin an der Universität Aberdeen und schloss sein Studium mit Auszeichnung ab. Im Anschluss studierte er Biochemie in Leipzig. Er wurde bereits 1903 zum Professor an der Western Reserve University in den USA berufen, wechselte aber später nach Toronto.

Macleod lieferte wichtige Arbeiten über die Zuckerkrankheit und den Kohlenhydratstoffwechsel. Für die Entdeckung des Insulins erhielt er 1923 gemeinsam mit Frederick G. Banting den Nobelpreis für Physiologie oder Medizin. Die Preisvergabe an ihn, und nicht an den wesentlich an der Entdeckung beteiligten Medizinstudenten Charles Best, war nicht unumstritten. Allerdings teilte er sein Preisgeld mit James Collip, dem Biochemiker, der für die Gewinnung des Insulin-Extraktes aus Pankreasgewebe verantwortlich war.
