Sulfhydryl

Die Thiolgruppe (alternativ Mercaptogruppe oder Sulfhydrylgruppe, –SH) ist eine funktionelle Gruppe, die der Hydroxylgruppe (–OH) analog aufgebaut ist. In ihr ist das stark elektronegative Sauerstoff-Atom durch das weniger elektronegative Schwefel-Atom ersetzt. Daher ist die Thiolgruppe weniger polar als die Hydroxylgruppe. Gleichzeitig wird das Proton am S-Atom leichter abgegeben als bei Alkoholen, weshalb die Thiolgruppe sau(r)er wirkt (analog zur steigenden Acidität H2O -> H2S). Molekülverbindungen, die analog zu den Alkoholen anstatt einer Hydroxygruppe eine Thiolgruppe tragen, nennt man Thioalkohole, Thiole oder (gemäß alter Nomenklatur) Mercaptane.

Organische Thiole weisen einen intensiven, unangenehmen Geruch auf (meist faulig). Aus diesem Grund muss man im Umgang mit Thiolen besondere Vorsicht walten lassen, denn diese Gerüche sind sehr "anhänglich".

Eine für den Bau vieler Proteine wichtige Aminosäure, das Cystein, trägt in ihrer Aminosäureseitenkette eine endständige Thiolgruppe, die für die Ausbildung von Disulfidbrückenbindungen entscheidend sind.
