Pyrophosphatasen katalysieren die Umwandlung von organischem Pyrophosphat zu Ortho-Phosphat. Es handelt sich um eine exergone Reaktion, womit andere, thermodynamisch ungünstige, biochemische Reaktionen ermöglicht werden, beispielsweise der Aufbau von Lipiden. Pyrophosphatasen findet man von der Bakterienzelle bis zum menschlichen Organismus in allen Organismen.

Thermus thermophilus besitzt eine für die Polymerase-Kettenreaktion (PCR) geeignete, temperaturbeständige Pyrophosphatase. Bei der PCR entsteht durch die Umsetzung von Desoxynukleotidtriphosphaten zu Desoxynukleotidmonophosphaten Pyrophosphat, das inhibihierend auf den Prozess wirken kann. Durch den Abbau von Pyrophosphat lässt sich die Effizienz und Ausbeute der PCR steigern.

Die Pyrophosphatasen aus Escherichia coli (ein Hexamer, also sechs Domänen) und aus menschlichen Zellen sowie Hefezellen (ein Dimer, also zwei Domänen) gehören zur Familie der Pyrophosphatasen 1. Pyrophosphatase 2 kommt ausschließlich in Prokaryoten vor.
